Daraxonrasib

Daraxonrasib is an investigational oral drug developed by Revolution Medicines that targets all three RAS proteins (KRAS, HRAS, and NRAS) to treat solid tumors. It works as a "molecular glue," binding to a chaperone protein (cyclophilin A) to form a tri-complex with RAS in its active (ON) state, effectively blocking the uncontrolled cancer growth driven by these mutations. This is highly significant because RAS mutations drive up to 30% of human cancers, including over 90% of pancreatic ductal adenocarcinomas (PDAC) and about 30% of non-small cell lung cancers (NSCLC). The drug received FDA breakthrough therapy designation in 2025, and its New Drug Application was accepted for review by the FDA in July 2026. Early clinical trials show a manageable safety profile—with common side effects like rash, diarrhea, and fatigue—making it a promising new candidate for patients with RAS-driven cancers.
